Immunohistochemical Localization of Irisin in Female BALB/c Mouse Genital Tissues

The number of studies demonstrating the roles of irisin, a myokine mainly produced by skeletal muscles, in female and male reproductive physiology has been increasing day by day. Therefore, the present study aimed to investigate the immunohistochemical localization of irisin in female genital organs. Oviduct, and uterus were rapidly excised from anesthetized female BALB/c mice (n = 6) and fixed in 10% buffered neutral formalin solution, followed by embedding in paraffin. Tissue sections of 5 µm thickness were obtained using a microtome and mounted on positively charged slides, after which they were stained using immunohistochemical methods. In the ovary, irisin-positive immunostaining was observed in all follicles except primordial ones, as well as in the germinal epithelium and corpus luteum. The staining intensity in granulosa cells increased in parallel with follicular maturation. The strongest irisin positivity was detected in luteal cells. In the oviduct and uterus, irisin-positive immunoreaction was detected in the surface epithelial cells, smooth muscle cells of the tunica muscularis layer, and endometrial gland epithelial cells. The findings obtained in this study regarding the expression pattern of irisin in female genital organs support the proposed beneficial roles of irisin in follicular development, uterine receptivity, and implantation.
