Abstract

Flow cytometry is an advanced analytical technique that enables rapid and multiparametric evaluation of the physical and physiological properties of microorganisms at the single-cell level. This review comprehensively addresses the technical infrastructure of flow cytometry and its expanding applications in food microbiology. The fundamental features of the instrument, its working principles, and analytical methods are explained. Flow cytometry allows for detailed analysis of cellular components and stands out in various applications in food microbiology, including the detection of pathogens, viability assessment of probiotics, monitoring of fermentation processes, and determination of microbial load in water sources. Compared to conventional culture-based methods, flow cytometry offers significant advantages such as faster results, the ability to distinguish between live and dead cells, and detailed insights into metabolic activity. Particularly in the analysis of l lactic acid bacteria (LAB), staining techniques based on membrane integrity and enzyme activity have yielded meaningful results. However, certain limitations arise from sample preparation, staining protocols, and the technical requirements of instrument operation. This study highlights both the strengths and limitations of the method and evaluates its current role in food microbiology in light of scientific evidence.
