Investigation of the Anticancer Effects on HeLa (Cervical Adenocarcinoma) Cell Line and the DNA Protective Activities of Medicinal Weed Plants from Ordu Province (Rubus canescens and Ficaria verna)

Abstract

This study investigates the protective potential of ethanolic extracts from medicinal weed plants from Ordu Province, Rubus canescens and Ficaria verna, against DNA damage (Compared to positive control Trolox) and their anticancer activity in HeLa (Cervix adenocarcinoma) cells. The extracts were prepared at varying concentrations, and their anticancer effects were evaluated using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay in HeLa cells. The ability of these extracts to prevent hydroxyl radical-induced damage to pBR322 plasmid DNA was also examined. MTT assay results showed that cell viability decreased in a dose-dependent manner when treated with different concentrations (6.125, 3.063, 1.531, 0.766, 0.383, 0.191, 0.096 mg/mL) of the extracts. The approximate IC50 values for R. canescens and F. verna extract at 24 hours were determined to be 2.23 and 0.26 mg/mL, respectively. At 48 hours, these values were 0.83 and 0.17 mg/mL, respectively. Furthermore, the extracts at various concentrations (40, 20, 10, and 5 mg/mL) demonstrated DNA protective activity comparable to Trolox in preventing hydroxyl radical-induced pBR322 plasmid DNA damage. These findings suggest that the ethanolic extracts of R. canescens and F. verna may serve as potential candidates for preventing DNA damage-related diseases due to their protective effects on plasmid DNA and controlled antiproliferative activities. However, further advanced cellular and in vivo studies are required to confirm these effects.
